Report 30th Month Project and Steering Committee Meeting FUCOMYR - Project QLKT-2001-02044

 

Project: Novel tools for developing Fusarium resistant and toxin free wheat for Europe.

Program of Day 1

1. Introduction

Welcome by PR; Appointing of PR and US as chairperson and secretary, respectively.

2. Approve of last meeting minutes

Last meeting minutes were unanimously approved.

 

3. Matters arising from last meeting

The following matters arose from last meeting:

a) Feedback concerning the successful financial transfer was missing,

b) we request from the financial officer to brake down the retained amount (to recover the initial advance) partner wise.

c) WP1: No limitations for the deliverables and milestones due in year 3 are foreseen, delays from year 2 will be corrected in year 3. No limitations for the deliverables and milestones due in year 3 are foreseen.

d) WP2: Maps will be refined in the coming season by adding additional SSR markers. High-resolution mapping populations are now advanced to F6-generation. Genotyping is underway, phenotyping will start in 2004 as planned.

e) WP3: Some changes in the ambitious subtask 2 and 3 are suggested in this work package due to recent developments within and outside the group of researchers. The Deliverables and milestones due to date are generally achieved, but have implications for, and suggest that modifications be made to, future work and milestones.

f) Finances not yet received (due date March 31, 2004, R. Hardwick will inquire about it) g) Deadline for reporting remains tight (therefore 1 month postponement from the of the scientific officer approved.

h) All should submit their presentation given at the meeting to allow follow up by each partner and co-worker in password protected area of our FUCOMYR website

6.    Detailed discussion of all work packages

 

WP1

After a detailed presentation of screening data from 2002 and 2003 the status of progress in WP1 has been reported by AM and ML (ST1, ST2, ST3, ST4).AM suggests to obtain additional FDK data for other populations as well. Based on one season data AM determined strong influence of plant height and anthesis on FHB severity. PR suggests additional analysis (i.e. rank correlations) and HB to include the 2004 data in the analysis. PN suggests a better phenotyping and resistance definition to distinguish between pleiotropie and linkage. FHB screening data from nurseries of P2, P3 and P4 are similar, while results between Szeged and Tulln differ more. Therefore for a subset of genotypes the inoculation methods of P1 and P5 will be compared in Tulln and Szeged in 2004. ML presents the efficiency of the 5A QTL against a number of Fusarium species. He observed that F. sporotrichioides does not cause high FDK values in difference to other Fusarium species tested. No limitations for the deliverables and milestones due in year 3 are foreseen. WP2

An overview about the progress within the work package was given.

Frontana/Remus QTL mapping: At IFA phenotyping and genotyping is completed. At Szeged (H) one more season of resistance testing is planned. Arina/Riband QTL mapping: Genotyping was refined with additional SSR markers by SURL. Phenotyping will be repeated on three sites (UK, A. H) in 2004. High-resolution mapping: Genotyping of  populations is underway at IFA. Plants are now ripening in the greenhouse of IFA, harvesting will start soon. Phenotyping on selected recombinants will start in 2004 as planned. Concerning the 5A QTL we are not sure yet if we are dealing with several linked genes or a single gene. Maps will be refined in the coming season by adding additional published SSR markers, which became available recently. We have to answer how to deal with the flanking marker and to determine, if the QTL is situated in a low combination area of the chromosome or elsewhere. Genotyping of screening nursery: Additional SSR markers have been analyzed on 96 lines of the screening nursery by SURL. Data are available at SURL for genetic similarity analysis, haplotype analysis and marker trait association analysis.For breeding applications there is the potential to combine the resistance coming from Frontana with resistance from Asian sources.We anticipate that the scientific activities as well as deliverables and milestones of WP 2 within the third project year can be achieved. WP3

FD, GA and PN reported about progress in WP3. The approved adjustments can be viewed in the password protected area of the FUCOMYR website. ST1: In the detached leaf assay correlations between latent period and FHB in commercial lines are functional. The SE microscopy work is underway. The Mycotoxin part of the detached leaf assays remains to be done until the end of the third project year. ST2: Full-length clones were obtained from a cv. Frontana cDNA library and we are currently trying to sequence these clones. We have sequenced two genes and are currently sequencing three more such genes. An analysis of the sequence data will be presented in the sixth report. Expression of some transcripts is higher in F. culmorum-inoculated cv. CM 82036 than in cv. Remus heads. PN reported, that the DH population was tested over two years for AUDPC, RSW, DON, FDNA. The 6B QTL was significant for several traits in both years. Populations were sown for new test in poly-tunnel and population was sent to Austria and Hungary for field testing. Concerning the ribosomal protein L3, allele-specific primers for 5A and 5D are used, and for Group 4 new primers developed. Some specific PDR sequences are generally increased in response to DON in FHB resistant and susceptible varieties. The accumulation of DON was reduced in grain extracts of some accessions of the mapping populations CM82036/Remus.K. Ansari, the Ph.D. student of FD will work for few month in GA laboratory in Austria continuing his work with candidate resistance genes.  GA reported that work has begun on the wheat cDNA library for yeast expression studies.

The Deliverables and milestones are achieved, and the approved adjustments will be met. The presentations of the WP3 results will be accessible in the password protected area of the FUCOMYR website.

7.    Technological implementation plan

The information based on the web based version of eTIP due at the end of the project was presented. Besides tasks under sole responsibility of the project coordinator, several points have to be addressed in a joint approach. Further, each partner has to fill out its own questionnaire towards the end of the project. Please view for details the management presentation.

 

8.    Consortium agreement

The part concerning the right to Veto decisions of the consortium will be removed from the agreement. If all partner completed the part of the pre-existing knowledge, the CA will be distributed to each partner ready to be signed.

 

9.    Other motions

Views, ideas, concerns and opportunities regarding the participation of our consortium within the 6th EU-Framework program were exchanged.

 

10.  Any other business

The project website was accessed – now located at the BOKU server - within the last three months 1385 times. Comments for any improvements were invited. A project flyer on glossy paper was not considered as necessary.

 

11.  Next meeting

The next meeting is the 36th month meeting in Hungary, where the participation of all project partner is required. September 27th and 28th, 2004 has been set as meeting date. It has been suggested to participate in the joint meeting of the US Wheat and Barley Scab initiative together with the European Fusarium Seminar in December 2004. The opinion to move the 36th month meeting in Norwich from February 2005 to Orlando, Florida by 2 month and use partly resource available for the steering committee meeting was discussed. The position of each partner will be requested to safeguard fairness within the consortium.

 

12.  Laboratory tour

The laboratory tour covered greenhouses and laboratories, where phytopathological and molecular methods relevant for the project were presented.
